The weather was perfect for this qualifying session, with 25° for the air temperature and 36° for that of the track. On the other hand, the wind was quite violent in certain turns. The reference time was the 1'42.413 established in FP3 by Aron Canet.

Romano Fenati was the first to set a significant time in 1'42.877. Jorge Martin improved this performance in 1'42.775, 0.01 ahead of Gabriel Rodrigo. The pace was already very fast at the start of the session. The first groups had formed and the aspiration was in full force, despite the modest length of the straight.

Rodrigo progressed in 1'42.485, then Fabio di Giannantonio took first place in 1'42.425, hoping to obtain his first GP pole position. He preceded Rodrigo by 0.06, himself 0.08 ahead of Aron Canet. Jules Danilo was twenty-eighth at 2.1, not looking much more comfortable than during free practice sessions (where he experienced electrical problems).

All the drivers returned to the pits with 25 minutes to go before the checkered flag, and Romano Fenati was the first to leave, all alone with no one glued to his rear wheel. He climbed into third position, 0.068 behind Fabio di Giannantonio and 0.008 behind Gabi Rodrigo. Aron Canet fell quite heavily on his lap exiting the pits, without seriousness. He was then in eighth position.

Nicolo Bulega moved up to second place, 0.022 behind di Giannantonio, and 0.026 ahead of Fenati. Three Italians then occupied the provisional first line at Misano. Jules Danilo fell without seriousness in the second turn.

With 15 minutes to go, Fabio di Giannantonio improved to 1'42.391, then returned to the pits like the entire field, while Aron Canet was taken to the medical center for inspection. Bulega's second time was canceled for exceeding the track limits, and the VR46 driver found himself eleventh.

The first drivers came out of the pits with less than five minutes to go. Aron Canet's motorcycle was not yet repaired. Everything was going to be decided at the end of the session on a single timed lap. Canet managed to leave just in time.

Pole position ultimately went to Enea Bastianini ahead of Jorge Martin, Joan Mir, Fabio di Giannantonio and Romano Fenati. Jules Danilo, due to his fall, only qualified in thirty-first position. A clearly difficult weekend for the Frenchman.
