It was on very wet asphalt that Marc Marquez achieved the best time of the Sunday practice session, ahead of Danilo Petrucci, Loris Baz, Jorge Lorenzo and Cal Crutchlow.

Due to a morning shower, the track was wet and numerous crashes took place during the warm-up in the Moto3 and Moto2 categories. However, it was useful for the MotoGP riders to test the track in these wet conditions, because the weather forecast predicted rain for the beginning of the afternoon, therefore for the start of the MotoGP race at 14 p.m.
Free practice took place entirely in the dry. Since the Misano surface was redone at the start of 2015, only the 2015 MotoGP race had taken place on a wet track, with Marc Marquez winning ahead of Bradley Smith, Scott Redding and Loris Baz.
At the start of the session, Jorge Lorenzo and Loris Baz came into contact without consequence, other than a little irritation.

This in no way prevented Loris from setting the best time shortly after, 13 minutes from the checkered flag in 1'51.380. Marc Marquez improved to 1'49.985. Then Baz regained the advantage in 1'49.420. He preceded Danilo Petrucci by 0.471. Sam Lowes was a surprising fifth, 1.5 behind Loris.
Petrucci took command midway through the session in 1'49.164, ahead of Loris Baz, Marc Marquez and Sam Lowes. Dani Pedrosa fell heavily, but not seriously.

Dani left shortly after on his second motorbike. His teammate Marc Marquez took the lead in 1'48.705. He was ahead of Danilo Petrucci and Loris Baz. Marquez fell during the last laps, without seriousness. He maintained the best time ahead of Danilo Petrucci, Loris Baz, Jorge Lorenzo and Cal Crutchlow.
